French term

HMW de grande taille

French to English Medical Medical (general) clinical trial
Hello out there!

This is part of a request for additional information regarding a clinical trial.

Concernant la substance active:
L'origine des HMW de grande taille pour le lot de substance active du Procédé X présente une polydispersité plus importante que le lot du Procédé Y

I was the thinking "high molecular weight" but then there should be something that that is modifying. Am I on the wrong track?

I would appreciate your thoughts!

Thank you.

Anne Schulz Dec 30, 2017:
Thanks, Sue, that's right, polydispersité suggests molecular weight distribution. Then again, it could be size distribution as well (I am familiar with this term in the context of aerosol particles for inhalation) and have to do with the "grande taille". Plus, grammatically, the "origin" is what seems to be "polydisperse" in this phrase. Nothing wrong with assuming "high molecular weight", probably, in particular if Denise has found other English acronyms in the text. Yet reading authors' thoughts and silently cleaning up semi-correct semantical messes sends the wrong message, IMO (and can be dangerous in terms of liability, as I recently experienced).
Sue Davis Dec 29, 2017:
@Anne. I'm afraid I have to disagree@ as the term polydispersity seems to relate to molecular weight distribution http://pubs.acs.org/doi/abs/10.1021/cm048594i?journalCode=cm... I think HMW most likely = high molecular weight.
Denise Pavao (asker) Dec 29, 2017:
Thanks everyone! This documents actually does include several other English acronyms. Also, I've now come across documents in which HMWs is used as a noun (e.g. "As by size separation HMWs could not be baseline separated perfectly, a possible influence of the peak overlay was considered. " https://edoc.ub.uni-muenchen.de/13874/1/Printz_Miriam.pdf, and "For instance, measurements on acidic pH-stressed mAbs, as reported by B. Veestergaard,
22 show a significant loss of monomers while denatured oligomers and
HMWS become predominant" https://hal-ens.archives-ouvertes.fr/hal-01510479/document.)
Anne Schulz Dec 29, 2017:
Ask the client :-) As Drmanu49 pointed out on a previous occasion, the French are not really inclined to use English acronyms. Plus, as you pointed out, the thing to be qualified is missing if HMW is a qualifier like high-molecular weight. Plus, as Drmanu49 pointed out, "de grande taille" is redundant if HMW stands for high molecular weight. --> Chances are that HMW is not "high molecular weight", but some (non-standard?) French abbreviation/acronym, and unless your text provides further clues on what it might be, there is little hope to guess the right answer.
Drmanu49 Dec 28, 2017:
Liz, don't forget that this is also qualified by "de grande taille".

The dispersity index, or formerly polydispersity index (PDI), or heterogeneity index, or simply dispersity (Đ), is a measure of the distribution of molecular mass in a given polymer sample. Đ (PDI) of a polymer is calculated: , where is the weight average molecular weight and is the number average molecular weight.

Hi, there are various nouns that could follow HMW...doesn't the clinical trial document give more clues?
Drmanu49 Dec 28, 2017:
I think it is proteins or molecules that is missing nervertheless de grande taille seems redundent with HMW

Disaggregation of high-molecular weight species during downstream processing to recover functional monomer.
Xu X1, Didio DM, Leister KJ, Ghose S.
Author information
Abstract
The use of chaotropic agents to recover functional monomeric material was investigated for the downstream purification of an Fc-fusion protein containing high levels of high-molecular weight (HMW) species. In batch studies, chaotropic agents irreversibly disaggregated a majority of the aggregated protein. An integrated processing mode, termed as on-column disaggregation, was developed in which the protein was captured on Protein A chromatography and then a chaotropic agent was used to simultaneously elute the bound protein and disaggregate the HMW species. On-column disaggregation process resulted in protein recoveries of >95% and aggregation reduction of approximately 50%. Analytical results are presented showing that the recovered monomeric material was comparable to the reference protein in biochemical, biophysical, and pharmacokinetic properties. The kinetic and molecular mechanisms governing protein aggregation and disaggregation will also be elucidated. For the Fc-fusion protein studied here, incorporation of the disaggregation strategy in both batch and on-column modes led to an increase of >10% in overall downstream yield.

Peer comment(s):

agree Helen Genevier : HMW species seems a great solution to me, as there are so many possibilities besides protein and DNA. I'd precede it by "large" in case the author is referring to a subset of HMW species./You too, Sue :-)
